4-Dimethylaminopyridine (DMAP) is a highly efficient nucleophilic acylation catalyst. The electron-donating dimethylamino group resonates with the pyridine ring, strongly activating the nitrogen atom of the ring for nucleophilic substitution. This significantly promotes the acylation and esterification of alcohols, amines and acids, including substrates with high steric hindrance or low reactivity. Its catalytic activity is approximately 10⁴–10⁶ times higher than that of pyridine. Acyl transfer is a fundamental reaction in both natural processes and organic synthesis, and chiral DMAP is widely used as a catalyst for asymmetric acyl transfer.
